CAR products from novel sources: a new avenue for the breakthrough in cancer immunotherapy.

Chimeric antigen receptor (CAR) T cell therapy has transformed cancer immunotherapy. However, significant challenges limit its application beyond B cell-driven malignancies, including limited clinical efficacy, high toxicity, and complex autologous cell product manufacturing. Despite efforts to improve CAR T cell therapy outcomes, there is a growing interest in utilizing alternative immune cells to develop CAR cells. These immune cells offer several advantages, such as major histocompatibility complex (MHC)-independent function, tumor microenvironment (TME) modulation, and increased tissue infiltration capabilities. Currently, CAR products from various T cell subtypes, innate immune cells, hematopoietic progenitor cells, and even exosomes are being explored. These CAR products often show enhanced antitumor efficacy, diminished toxicity, and superior tumor penetration. With these benefits in mind, numerous clinical trials are underway to access the potential of these innovative CAR cells. This review aims to thoroughly examine the advantages, challenges, and existing insights on these new CAR products in cancer treatment.

Abstract 4935: Cannabidiol and cannabis extract protect cells against platinum-based chemotherapy via cellular uptake

Abstract Cannabinoids including cannabidiol belong between constituents of cannabis sp. (marijuana plant). Purified or as a part of cannabis products, the compounds are frequently used by cancer patients to attenuate chemotherapy-induced side effects such as pain, nausea, and vomiting. The presence of cannabis products on the public market expands each year while the illegality status weakens in an increasing number of countries. U.S. Food and Drug Administration has approved several cannabis-based drugs for seizures induced by Lennox-Gastaut syndrome, Dravet syndrome, or tuberous sclerosis complex, also for the lost appetite of AIDS patients, and chemotherapy-induced nausea and vomiting. Oncological patients are further and strongly motivated to take the drugs in combination with chemotherapy by numerous reports demonstrating the anticancer effects of cannabinoids. Nevertheless, here we present unsettling data that cannabis extract together with CBD negatively interferes with worldwide approved platinum-based drugs cisplatin, carboplatin, and oxaliplatin. The phenomenon is proven across three cellular models including primary cells. Even low concentrations of cannabidiol or cannabis extract reduced the toxicity of platinum-based drugs using long-term colony forming assay and/or short-term crystal violet assay. Increased resistance of cells was also accompanied by decreased cellular stress as evaluated by quantitative immunofluorescence microscopy and immunoblotting. Most importantly, concomitant cannabidiol and cisplatin exposure results in the decreased presence of cisplatin-modified DNA. Following the information, we determined platinum content in cells by ICP-MS. The experiment uncovered significantly decreased intracellular platinum content indicating an effect on cellular transport. Furthermore, the mechanism is translational independent as shown by the experiment with cycloheximide. This study offers crucial information that patients and physicians should take into account regarding possible interactions between platinum-based drugs and cannabis-based products. Furthermore, the study on the contradictory effects of cannabis products in cancer treatment may be motivated by this project. Potential, Limitations and Risks of Cannabis-Derived Products in Cancer Treatment.

The application of cannabis products in oncology receives interest, especially from patients. Despite the plethora of research data available, the added value in curative or palliative cancer care and the possible risks involved are insufficiently proven and therefore a matter of debate. We aim to give a recommendation on the position of cannabis products in clinical oncology by assessing recent literature. Various types of cannabis products, characteristics, quality and pharmacology are discussed. Standardisation is essential for reliable and reproducible quality. The oromucosal/sublingual route of administration is preferred over inhalation and drinking tea. Cannabinoids may inhibit efflux transporters and drug-metabolising enzymes, possibly inducing pharmacokinetic interactions with anticancer drugs being substrates for these proteins. This may enhance the cytostatic effect and/or drug-related adverse effects. Reversely, it may enable dose reduction. Similar interactions are likely with drugs used for symptom management treating pain, nausea, vomiting and anorexia. Cannabis products are usually well tolerated and may improve the quality of life of patients with cancer (although not unambiguously proven). The combination with immunotherapy seems undesirable because of the immunosuppressive action of cannabinoids. Further clinical research is warranted to scientifically support (refraining from) using cannabis products in patients with cancer.

The efficacy of applying some plants and herbs in cancer therapy for humans and animals – A comprehensive review

Abstract Cancer is a challenging ailment and represents the main reason for death worldwide for humans and animals. Although great developments have hindered cancer progression, several adverse effects are associated with modern chemotherapy. Natural remedies, such as the usage of medicinal plant or their products in cancer treatment, may decrease prejudicial side properties. Recently, the modern research scheme and innovative screening practices for herbs or plants have enabled phytochemical discovery for the prevention and treatment of cancer. This criticism highlights herbs such as acacia, basil, black seeds, cedar, castus, ficus, garlic, ginger, indigo, onion, pomegranate, quince, and thyme, promising anticancer effects. The present review also revealed the mode of action of each herb as anticancer effects at level in vitro and in vivo studies. The item also totalizes the vital mechanisms and signaling molecules involved in preventing cancer diseases. This will fill the investigation gap in the exploration of using natural molecules and encourage researchers in clinical trials of anticancer agents from herbs for humans and animals.

Clinical Research Evidence Supporting Administration and Dosing Recommendations of Medicinal Cannabis as Analgesic in Cancer Patients

The analgesic potential of Cannabis sativa L.-based medicinal cannabis products for treatment of cancer associated chronic pains has gained increased interest in recent years. To ensure a controlled distribution of these products and investigate their therapeutic potential, several countries have established so-called pilot trials. Many doctors, however, are hesitant to prescribe medicinal cannabis primarily due to lack of research evidence regarding the products' efficacy, safety and thus questionable dosing guidelines. This review aims to elucidate clinical research supporting administration of medicinal cannabis in cancer patients for analgesic purposes. The cannabinoids' effects on the endocannabinoid system (ECS) and its implication in pain regulation is included to illustrate the complexity related to this research field. Published clinical studies on medicinal cannabis primarily consist of observational studies and only one pilot randomized controlled trial (RCT), where more RCTs exist on the cannabis-based product, Sativex® (GW Pharma Ltd., Cambridge, UK). The studies indicate analgesic potential, however non-significantly, for most patients and with acceptable safety profile. Summarizing, high-quality RCTs are scarce in this research field, and the limitations of the observational studies complicates interpretation of clinical outcomes. Despite discrepancy among the studies, they do show indications for administration and dosing regimens providing analgesic effects for some cancer patients.

The Impact of Oxidative Stress and AKT Pathway on Cancer Cell Functions and Its Application to Natural Products

Oxidative stress and AKT serine-threonine kinase (AKT) are responsible for regulating several cell functions of cancer cells. Several natural products modulate both oxidative stress and AKT for anticancer effects. However, the impact of natural product-modulating oxidative stress and AKT on cell functions lacks systemic understanding. Notably, the contribution of regulating cell functions by AKT downstream effectors is not yet well integrated. This review explores the role of oxidative stress and AKT pathway (AKT/AKT effectors) on ten cell functions, including apoptosis, autophagy, endoplasmic reticulum stress, mitochondrial morphogenesis, ferroptosis, necroptosis, DNA damage response, senescence, migration, and cell-cycle progression. The impact of oxidative stress and AKT are connected to these cell functions through cell function mediators. Moreover, the AKT effectors related to cell functions are integrated. Based on this rationale, natural products with the modulating abilities for oxidative stress and AKT pathway exhibit the potential to regulate these cell functions, but some were rarely reported, particularly for AKT effectors. This review sheds light on understanding the roles of oxidative stress and AKT pathway in regulating cell functions, providing future directions for natural products in cancer treatment.

Cytotoxic activity and antioxidant effects of Origanum onites essential oil and its two major contents, carvacrol and p-cymene on human colorectal (HCT116) and hepatocelluler carcinoma (HepG2) cell lines

ABSTRACT Origanum onites L. (O. onites) is one of the cultivated Origanum species in Turkey. The aim of this study was to determine the cytotoxicity and cellular antioxidant effects of O. onites essential oil, carvacrol and p-cymene on HCT116 and HepG2 cells. O. onites essential oil components were analyzed by GC/MS and GC/FID. Cytotoxicity was analyzed via the MTT assay in HCT116 and HepG2 cells. Cellular antioxidant activities of O. onites essential oil, carvacrol and p-cymene were determined with DCFH-DA method. GC/MS analysis identified carvacrol (%78.4) as the main constituent of O. onites essential oil. 400 µg/ml concentration of O. onites essential oil, carvacrol and p-cymene were found to be active against HCT116 and HepG2 cells. Also, carvacrol showed higher cellular antioxidant activity than O. onites essential oil and p-cymene. We conclude that O. onites essential oil and carvacrol may be a candidate for pharmaceutical products in treatment of cancer.

A Review of Himalayan Medicinal Plants against Cancer

Background: Plants contributed numerous novel compounds for prophylactic and curative medicine to modern science. They are an important source of natural agents used in various pharmaceutical industries. Himalayan plants are abundant in various secondary metabolites, such as anthraquinones, flavonoids, tannins, alkaloids, and terpenes. The Himalayan plants are grown in high altitudes and have around 6500 years old history as traditional medicines. Aim: This review article systematically presents information on Himalayan medicinal plants having anti-cancer potentials. Methodology: Around 160 articles were reviewed using online search engines, like PubMed, Scopus, Google Scholar, Web of Science, and floras of different Himalayan countries. Results: An attempt has been made to review anti-cancer plants and active constituents isolated from several anti-cancerous plants of Himalayan regions. Eighty-three anti-cancer plants are reported in this review, and a total of twenty-two active chemical constituents, including phenolic compounds, glycosides, terpenoids, and alkaloids from the plants, were reported active against various cancer cell lines. Conclusion: Several synthetic agents are used to cure cancer, but many undesired side effects occur during chemotherapy. Hence, the research is going on to investigate natural therapies, such as the use of plant-derived products in cancer treatment. They may reduce adverse side effects.

Tumor-derived extracellular vesicles as messengers of natural products in cancer treatment.

Extracellular vesicles (EVs) are kinds of two-layer vesicles secreted by cells. They play significant roles in mediating component exchange between cells, signal transduction, and pathological development. Among them, the tumor-derived EVs (TDEVs) are found related to the tumor microenvironment and cancer development. TDEVs can be designed as a natural drug carrier with high tumor targeting and permeability. In recent years, drug delivery systems (DDS) based on TDEVs for cancer treatments have received considerable attention. In this review, the biological characteristics of TDEVs are introduced, especially the effect on the tumor. Furthermore, the various approaches to constructing DDS based on TDEVs are summarized. Then we listed examples of TDEVs successfully constructing treatment systems. The use of chemical drugs, biological drugs, and engineered drugs as encapsulated drugs are respectively introduced, particularly the application progress of active ingredients in traditional Chinese medicine. Finally, this article introduces the latest clinical research progress, especially the marketed preparations and challenges of clinical application of TDEVs.

Cytotoxic Effects of Eupatorium cannabinum on MCF-7 Human Breast Cancer Cell Line

Background and objectives: Cancer is known to be the second cause of death around the world. The most prevalent cancer among women is breast cancer. Use of plant-derived products in cancer treatment may reduce adverse side effects. Extensive research around the world has led to the discovery of herbal compounds that can be used to treat some types of cancer. According to previous studies, the methanol extract of Eupatorium cannabinum has shown cytotoxic effects in some cancer cell lines. In the present study, bioassay guided fractionation and isolation was conducted on E. cannabinum to evaluate the cytotoxic activityin MCF-7 breast cancer cell line. Methods: The Extraction from theaerial parts was performed by maceration method. Isolation and purification of the extracts were performed by column chromatography. The cytotoxic activity of different extracts of E. cannabinum was evaluated against MCF-7 cell line by MTT assay and a compound was isolated according to bioassay guided fractionation. The cytotoxic activity and apoptotic property of the isolated compound was determined. Results: The chloroform extract was the most active one with IC50 of 21.39±3.24 μg/mL followed by the n-hexane and methanol extracts with IC50values of 60.23±2.16 μg/mL and 81.74±3.41 μg/mL, respectively. IC50 of subfractions (1-6) from the chloroform extract were 60.83±2.56 μg/mL, 58.93±2.73 μg/mL, 37.5±3.65 μg/mL, 7.86±1.34 μg/mL, 10.61±2.34 μg/mL and 13.77±4.17 μg/mL, respectively. Eucannabinolide, a sesquiterpene lactone, was isolated from the chloroform extract according to bioassay guided fractionation. Its IC50 was found to be 13±2.45 μg/mL. Eucannabinolide induced 46.91% apoptosis at concentration of 13 μg/mL in MCF-7 cell line in Annexin V/PI assay. Conclusion: Eucannabinolideis a promising candidate for further breast cancer drug discovery studies.

AKT/GSK-3 Pathway Targeting; Botanicals and Bioactive Compounds with Anticancer Activities.

In the late 20th century, the leading role of signaling pathways in various cancers is revealed via some genome's systematic investigations. The Akt/GSK-3 signaling pathway is one of the critical signaling pathways dysregulated in numerous human cancers. The Akt cascade acts in the cancer process by regulating apoptosis, cell cycle, metabolism, and cells' longevity. The GSK-3 is downstream of Akt, which has an opposite role in cancer progression. Attending to the importance of the Akt/GSK-3 pathway in cancer progression and the positive result of natural products in cancer treatment, this research is designed to review effective herbal medicines in one of the involvement critical signal pathways of cancer for developing novel anticancer drugs. Keywords "plant", "natural", "cancer", "AKT", and "GSK" were searched through the "Scopus" and "Google scholar" databases up to 30th August 2020. Papers linking to pharmacology, toxicology, and pharmaceutics were collected and discussed. The Akt/GSK-3 signaling pathway plays a prominent role in cancer. Although the effect of GSK-3 in cancer cells is depended on the type and contents of cells, the inhibition of Akt/GSK-3 mostly is led to three primary outcomes in cancer cells, including (1) apoptotic activity and autophagy induction, (2) anti-proliferative and growth inhibitory effects, and (3) anti-metastatic and angiogenesis effects. As the tendency to use natural products increases, we gathered 64 plants or bioactive components with the anticancer activity via the Akt/GSK-3 signaling pathway. Since most of these investigations have been conducted on cell lines, these plants can be the right candidate to be evaluated in human trials.

Research progress of natural plant products inducing apoptosis of tumor stem cells

Compared to general cancer cells, cancer stem cells are stronger and more difficult to be killed by drugs. Therefore, traditional treatment methods have low efficacy according to cancer stem cells. In essence, one of the main causes of cancer survival, proliferation, transfer, and recurrence is the presence of cancer stem cells. In recent years, a large number of studies have demonstrated that natural plant products have powerful in killing cancer cells and have various structures, which possess low toxicity, multi-target regulation advantages in inducing cancer stem cells to apoptosis. Therefore, this provides new ideas for developing anti-cancer drugs by inducing cancer stem cell apoptosis by natural plant products. In this article, we will introduce the apoptotic mechanism of tumor stem cells, the importance of eliminating tumor stem cells, and the benefits of natural plant products in cancer treatment. In addition, this paper reviewed nine categories of chemicals in natural plant products that induce apoptosis of tumor stem cells and their mechanisms of action. We also summarized the mechanism of natural plant products inducing apoptosis of tumor stem cells. This review provides an important reference for the personalized treatment of cancer.

Anticancer Potential of Aguerin B, a Sesquiterpene Lactone Isolated from Centaurea behen in Metastatic Breast Cancer Cells.

Breast carcinoma is a malignant disease that represents the most common non-skin malignancy and a chief reason of cancer death in women. Large interest is growing in the use of natural products for cancer treatment, especially with goal of suppression angiogenesis, tumor cell growth, motility, as well as invasion and metastasis with low/no toxicity. It is evident from recent patents on the anticancer properties of sesquiterpene lactones such as parthenolide. In this study, using MDA-MB-231 cells of a human breast adenocarcinoma, the effects of aguerin B, as a natural sesquiterpene lactone, has been evaluated, in terms of the expression of metastatic-related genes (Pak-1, Rac-1 and HIF-1α). Cytotoxicity of aguerin B was tested toward MDA-MB-231 breast tumor cells using MTT. Scratch assay was accomplished to evaluate the tumor cell invasion. To understand the underlying molecular basis, the mRNA expressions were evaluated by real time PCR. It was found that aguerin B significantly inhibited human breast cancer cell growth in vitro (IC50 = 2μg/mL) and this effect was accompanied with a persuasive suppression on metastasis. Our results showed that aguerin B in IC50 concentration down-regulated Rac-1, Pak-1, Hif-1α and Zeb-1 transcriptional levels. Taken together, this study demonstrated that aguerin B possessed potential anti-metastatic effect, suggesting that it may consider as a potential multi target bio compound for treatment of breast metastatic carcinoma.

IN-VITRO SCREENING OF SOME PLANT EXTRACTS FOR THEIR POTENTIAL ANTICANCER ACTIVITY

Background: Natural products have been shown to be reliable sources of anticancer medicines although there is still a consistent demand for new therapeutic natural products for cancer treatment with minimal side-effects.Materials and Methods: In this study, six plant extracts (Grevillea robusta; Euphorbia millii; Euphorbia royleana; Aloe grandidentata; Bauhinia corniculate; and Cassia fistula) were screened for the presence of phytochemical metabolites as saponins, tannins, cardiac glycosides, alkaloids, flavonoids, anthraquinones and sterols, using qualitative tests. Antiproliferative screening assay was performed on a panel of three cancer cell-lines (HepG-2, HCT-116 and MCF-7) using MTT assay, and cytotoxicity was determined using WI-38 human fibroblast cell- line.Results: Some plant extracts reduced cellular growth for the selected cancerous cell-lines. For example, E. royleana and A. grandidentata extracts reduced HepG-2 cellular growth with IC50 of 0.42 and 0.53μg/mL, respectively. Moreover, A. grandidentata and C. fistula reduced cellular growth of MCF-7 with IC50 of 0.37 and 0.67 μg/mL, respectively. Conclusion: E. royleana, A. grandidentata and C. fistula showed significant anti-proliferative activity against HepG-2 and MCF-7 cell-lines with non-cytotoxic nature. This suggests their potential role as anticancer agents against these types of cancer. The presence of flavonoids, sterols and anthraquinones may suggest their enhanced anti-proliferative activities. Therefore, this study has shed light on the possible use of these extracts as potential sources of natural products-based therapy for cancer.

The Current Use of Biomedical Cell Products for Cancer Treatment

Cancer remains one of the leading causes of death. Conventional treatment methods, including radiation and chemotherapy, have limited effectiveness. Therefore, the development of novel approaches to cancer treatment is an urgent challenge. Biomedical cell products (BMCPs) which include adoptive cell therapy (ACT) and dendritic cells vaccines (DCVs) are considered a promising area of research. The aim of the study was to review current ideas about the principles of BMCP therapy, as well as clinical experience with cell-based products used for cancer treatment. The paper summarises the results of clinical use of tumor-infiltrating lymphocytes (TIL-therapy), genetically modified T-cells that express tumour antigen-specific receptors (TCR/CAR T-therapy), as well as DCVs. The use of human immune cells genetically modified ex vivo is a novel and promising approach to cancer treatment. The main analysed ACT approaches which are based on the use of genetically modified T-lymphocytes have some benefits and drawbacks. The paper discusses the methods of BMCP production, provides data on the effectiveness of ACT and DCVs. It pays special attention to safety concerns associated with each treatment method, as well as to other factors limiting their clinical use. It is expected that the main areas of further research will be aimed at increasing BMCP efficacy and reducing adverse reactions.

Economic Analysis And Evaluation Of The Value Of The Parallel Export Of Medicinal Products For Treatment Of Oncological Diseases In Bulgaria, 2016 T. VEKOV 1 AND G. KOLEV2

The aim of the research is to analyze and assess the parallel export of medicines with annual sales of >3 million BGN for treatment of oncological diseases from Bulgaria in the year 2016. Risk assessment for the shortage of life-saving medicinal products. A documentary method and analysis was used to compare the quantity and value of oncological medicinal products sold to distributors in Bulgaria in 2016 and paid by the National Health Insurance Fund (NHIF), reported by two independent sources - IMS Health (IMS) and National Health Insurance Fund (NHIF). The study included products with annual sales of> 3 million BGN. The economic analysis carried out estimated the value of the parallel export of medical products for treatment of cancer from Bulgaria in 2016 at 43.2 million BGN. Products of 5 pharmaceutical manufacturers account for 87.2% of the value of the parallel export: Roche (52.8%) - bevacizumab, trastuzumab, rituximab , erlotinib, pertuzumab, vemurafenib; BMS (14.3%) - dasatinib; Bayer (9.2%) - sorafenib, Novartis (6.2%) - everolimus, nilotinib, pazopanib; Astellas (4.7%) – enzalutamide. Conclusions: This study does not reveal any evidence of either medical product shortage nor limited access to treatment for patients due to lack of access to drug therapy in Bulgaria

Medicinal and Economic Values of Forest Products in the Treatment of Cancer in Southwest Nigeria

Medicinal plants are used to address the twin problems of promoting sustainable livelihoods and treatment of numerous illnesses in Nigeria. The study examined the medicinal value of forest products in the treatment of cancer in South-west Nigeria. Primary data was obtained in a cross-section survey of 327 respondents comprising 127 Traditional Medicine Practitioners (TMPs), 100 Orthodox Medicine Practitioners (OMPs) and 100 respondents from the General Public drawn by multistage sampling technique from the study area. Interview schedule was used in collection of data on the effectiveness of forest products in cancer treatment. The result showed that seven species were identified belonging to seven different families; Rutaceae, Asteraceae, Anarcardiaceae, Annonaceae, Meliaceae, Guttiferaceae and Leguminaceae topped the TMPs priority list. Result of economic analysis shows minimal competition in the anti-cancer forest product market and a high level of monopoly with a Gini coefficient of 0.83. The rate of return on investment was 180.08% indicating that the TMPs were making profit. Five of the plants were tested against cancer cell lines MCF7 and Hs578T while Doxorubicin (a synthetic anticancer drug) was used as the control treatment. Three plants: Saccharum offinarum (Stem), Sucurinega virosa (Root) and Piper guineensii (Seed) produced no result; Garcinia kola (Bark) did not exhibit any anticancer effect even at a concentration of 10 μ1/m1 while only one plant species was effective against the cancer cell line at 1 μ1/m1. It is therefore concluded that forest products are effective in the treatment of cancer.

CYTOTOXICITY OF STEM EXTRACTS OF SELECTED CASSIA SPECIES AGAINST HELA AND BREAST CANCER CELL LINES IN VITRO

ABSTRACTObjective: The use of plants for medicinal remedies is an integral part of the Indian cultural life, and the traditional background of Indian medicineshows widespread use of plant products in cancer treatment. In this study, stem extracts of some selected Cassia species have been evaluated for theircytotoxic activities under in vitro conditions.Methods: The stems were shade dried at room temperature. The dried and coarsely powdered plant material were extracted with petroleum ether(60-80°C), chloroform, and ethanol using soxhlet apparatus. The cytotoxicity was evaluated by [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide] assay.Results: Stem extracts of three Cassia species, viz., Cassia glauca, Cassia Obtusifolia, and Cassia sophera have been evaluated for their cytotoxic activitieswith chloroform, ethanol and pet ether against HeLa and breast cancer cell lines. Among the three different solvents used at different concentrations,the chloroform extracts of all Cassia species exhibited maximum cytotoxicity (%) against both cell lines. The CTC50 values are revealed the cytotoxicpotential of C. glauca chloroform extracts against HeLa cell line and breast cancer cell lines with CTC50 values 180.00±3.0 and 146.67±0.5, respectively.Chloroform extracts of C. Obtusifolia and C. sophera showed maximum activity against HeLa (380.00±1.1 and 800.00±1.7, respectively) and breastcancer cell lines (310.00±1.1 and 633.33±0.6).Conclusion: The results of this study demonstrate the potent cytotoxic activity of chloroform extracts of stems of Cassia species against HeLa andBreast cancer cell lines.Keywords: Cassia, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide, Cytotoxicity, HeLa, Breast cancer.

The Efficacy of Some Comestible Natural Products in Treatment of Cancer

Despite the fact that cancer is primarily a preventable disease, recent statistics indicate cancer is the number one killer worldwide. Conventionally, cancer therapy includes surgery, radiation and drugs, separately or in combination. This modality of cancer treatment is still not successful. Therefore, the identification of new cytotoxic drugs with low or none side effects is of great interest worldwide. Since traditional medicines have been used for maintaining health, as well as in the prevention, therapy and remission of cancer, natural products can serve as chemopreventive and chemotherapeutic agent. Here, we describe the efficacy of 3 comestible natural products such as garlic, green tea and cruciferous vegetables in cancer therapy.
